Pipeline and animal health monitoring were conducted in the Department of Veterinary Medicine of the Faculty of Veterinary Science and Veterinary Medicine in Wuhan University (Hubei, China) and China Veterinary College in Wuhan, China. This study was carried out in accordance with the recommendations of the Pharmacoepidemiology of Veterinary Medicine (Ph. E. M. C) and the Animal Health Research Ethics Committee of Wuhan University, China, and was approved by the Ethics Committee of the Animal Health Research Ethics Committee of Wuhan University (Hubei, China). All animals were kept in a dark/light-dark cycle and had no known history of cardiovascular disease, hypersensitivity, or cardiovascular disorders. The animals were provided with food and water, and were allowed to acclimate to the new environment before the experiments began. The doxycycline and doxycycline hyclate tablets were administered orally (doxycycline 100 mg/kg, trihydrochloride 10 mg/kg, and doxycycline 100 mg/kg, respectively) and orally (doxycycline 100 mg/kg, trihydrochloride 10 mg/kg, and doxycycline 100 mg/kg, respectively) for 5 days. At the end of the 5 days of treatment period, the animals were euthanized and the blood samples for biochemical and histological analyses were obtained. The doxycycline and doxycycline hyclate tablets were separated by a polypropylene bottle and the blood samples were collected by an intravenous drip. The serum level of tetracycline was determined by high-performance liquid chromatography (HPLC) method. The tetracycline blood serum concentrations were obtained by using a commercially available kit (Bio-Rad, Hercules, CA, USA). The tetracycline concentration was measured using a C18 chromatograph (Agilent Technologies, Santa Clara, CA, USA). The concentration of doxycycline was determined using a C18-Chromatographic HPLC method with a detection wavelength of 210 nm. The tetracycline concentration was determined using a C18-Chromatographic HPLC method with a detection wavelength of 260 nm. All the experiments were performed in triplicate, and the data were expressed as the mean ± SD. The statistical analyses were performed using the Review Manager 4.3.3 software (Oxoid Inc., Missil, UK).

Samples were collected by the intravenous route, and samples were stored at -20 °C until analysis. The mean and the standard deviation of the mean were calculated for each animal. The concentration and tetracycline concentration of doxycycline and doxycycline hyclate were calculated using the standard curve and the standard concentration (S. D.) from the mean values of the concentration and the tetracycline concentration from the standard concentration of the drug.

The bioequivalence of doxycycline and doxycycline hyclate was evaluated by the following formula:

Eq. (1) is the concentration and tetracycline concentration of doxycycline and doxycycline hyclate tablets in each animal (in µg/mL).

In the following experiments, the mean and the standard deviation of the mean were calculated for each animal.

The pharmacokinetic profiles of doxycycline and doxycycline hyclate in dogs receiving different dosages of doxycycline, are shown in

    (2) is the concentration and tetracycline concentration of doxycycline and doxycycline hyclate tablets in each animal (in µg/mL).
